A transgenic animal model describes a model which has been genetically modified through the use of genetic engineering techniques. For several animal models PolyGene offers efficient (non-) random integration of your transgene.

At PolyGene, designed plasmids or BACs are injected in the pronucleus of the oocyte and integrate randomly or non-randomly into the genome. The main advantage of this technique is the short development time – it takes about 2 months. Level and pattern of the transgene expression can vary due to unknown copy number and integration site, which makes it necessary to analyze several independent transgenic lines.

BAC pronuclear microinjection
Bacterial artificial chromosomes (BACs) carry large cloned segments of native genomic DNA (100–300 kb). Consequently, they can carry entire mammalian genes including nearly all structural and cis acting regulatory elements. Because these elements are maintained in their native configuration, BAC transgenes are the ideal tool for delivering expression that is a precise recapitulation of the endogenous gene including cell type specificity, cell cycle, and mRNA regulation by microRNAs.

BACs from various genome resources are readily available and the development of recombineering technology has soared (Copeland et al., 2001; Hofemeister et al., 2011; Muyrers et al., 1999; Zhang et al., 1998). This strongly suggests BAC transgenesis is the preferred option for transgenesis in many systems.

However, BAC pronuclear injection generally is demanding and the integrity and copy number of the BAC transgene is difficult to control.
That’s why PolyGene offers a precise pronuclear microinjection method that relies on piggyBAC transposase mediated insertion of only full-length BACs into the mouse genome.

The transposase mediated BAC integration guarantees:

  1. Increased frequency of transgenic founders.
  2. Increased rate of intact, full-length, single copy integrations is increased.
