Transgenic Services

PolyGene provides a complete range of conventional transgenic services, which are offered on a modular basis. Projects are split up into milestones, each of which is payable in two installments (30% of the sub-project value upfront, and the remaining 70% upon completion and acceptance of the results).

Gene Cloning

Starting from the accession number, our molecular biology staff can reliably:

 construct an adequate probe, and screen and isolate the clone of your choice
   (e.g. from genomic BAC or cDNA libraries), and verify your clone inserts by
   Southern and/or PCR technology
 screen databases for publicly available clones (BAC, cDNA), and retrieve them
 generate sequences via total gene synthesis that are not otherwise available

in approximately 2 months time.



Vector Construction

State-of-the-art transgenic expression vectors can be constructed based on individual requirements, and on available sequences and base constructs. The service can comprise:

 assembly of standard expression cassettes (promoter and gene of interest)
 assembly of Cre-activatable transgenes
 inclusion of additional control elements, and construct sizes of up to 25 kb
 establishment of founder/offspring screening PCR screening procedures
 establishment of a quantification procedure for transgenes

and can be performed in 1-4 months (depending on complexity).



Inducible Systems

PolyGene disposes of several mutually independent models for inducible expression in transgenic animals, and can offer these free of rights. A transgenic inducible model can include:

 the design of an effector cassette, expressing the gene responsive to the
   inducing substance (antibiotic, acetaldehyde, hormone)
 the design of the responder cassette, with the gene of interest to be
   activated/suppressed
 generation of two different genotypes (effector, responder) for double-transgenic
   breedings, or
 assembly of the cassettes into one system and generation of a single transgenic
   line

These models can be developed in the course of a vector construction project, but are usually more complex and will require up to 6 months of development time

Pronucleus Injection

Transgenic animals are preferably generated on CB6F2, FVB/N, or C57Bl/6N mouse or Sprague Dawley rat genetic backgrounds. The service includes:

 preparation of the vector DNA and isolation of an appropriate fragment injection
   of zygotes,
 transfer into IVC-held foster mothers,
 maintenance, tagging and biopsying of offspring
 usually, in dependence of mouse strain and type of vector (plasmid, BAC), a
   minimum number of offspring is guaranteed
 optionally, guaranteed for founders can be given in connection with a PCR
   screening and verification step

within 2-4 months.
